L'objectif principal de notre groupe de recherche est d'étudier les mécanismes de transfert de molécules d’intérêt thérapeutique dans les cellules et les tissus par applications d’impulsions de champs électriques et de développer de nouveaux outils de délivrance, de ciblage et de diagnostic. Pour cela, nous adaptons et améliorons différents systèmes d’étude complémentaires à complexité croissante (liposomes, cellules en culture, sphéroïdes et petit animal) et intégrons divers outils d'imagerie pour visualiser et définir les phénomènes de perméabilisation membranaire et cellulaire au niveau moléculaire.

Nous développons une approche multidisciplinaire combinant la biologie cellulaire et la biophysique pour déterminer les mécanismes des perturbations membranaires induites par les modifications du potentiel transmembranaire, c'est-à-dire par la technique d'électroporation ou d'électroperméabilisation.

Dans ce contexte, et sur la base de nos travaux antérieurs, les principaux objectifs que nous avons poursuivis sont :

•    L’élucidation des mécanismes d'électrotransfert : nos travaux mettent en évidence les différences entre le transfert de petites molécules (bléomycine, cis-platinium), de petits oligonucléotides (siRNA, LNA), de protéines et d'ADN plasmidique et entre les mécanismes impliqués in vitro sur cellules en culture (modèles bidimensionnels et tridimensionnels) et in vivo sur tissus (modèle murin).

•    Le développement de nouveaux outils de délivrance, de ciblage et de diagnostic : nos résultats nous ont conduit à définir de nouveaux types de générateurs d'impulsions électriques et d'électrodes (adaptés pour l'étude in vitro et in vivo) et à adapter les systèmes d'imagerie (chambre à fenêtre dorsale, micro et macroscopie à fluorescence). De plus, nous développons d'autres approches d'administration de médicaments non viraux basées sur la lectine et nous utilisons des aptamères d'ADN qui se lient spécifiquement aux cellules cancéreuses. Cette approche a des intérêts pour le ciblage des tumeurs (administration de médicaments) et le diagnostic (imagerie des tumeurs solides). 

Nous travaillons en collaboration avec des laboratoires du monde entier dans le cadre de plusieurs réseaux internationaux (International Bioelectrics Consortium, COST Action TD1104 : réseau européen pour le développement de technologies et traitements basés sur l'électroporation et LIA-EBAM : International Associated Laboratory on Pulsed Electric Fields Applications in Biology and Medicine).
