Mamadou Daffé started his research activities by studying the lipids of Mycobacterium leprae in 1980 in the lab of Jean Asselineau, the discoverer of mycolic acids in 1950. He obtained his PhD in Biochemistry and was recruited by the CNRS as Associate Researcher in 1982. He obtained his Doctorat d’Etat in 1986 in Molecular and Cellular Biology, with a specialty in Pharmacology. Since then, his work focuses on the nature and organization of the mycobacterial cell envelope. During a post-doctoral stay (1990) at the Colorado State University (Fort Collins), in the lab of Pr Patrick J. Brennan, he deciphered the structure of the cell wall arabinogalactan and its links with mycolic acids and peptidoglycan. He became Research Director in 1993 at CNRS and the Principal Investigator of the team. Since then, he determined the chemical composition of the mycobacterial capsule (1994-1996). He founded the Department of “Molecular Mechanisms of Mycobacterial Infections” of IPBS, which he headed from 2000 to 2010.  In collaboration with the lab of Jacques Dubochet (laureate of the Nobel Prize of Chemistry, 2017), he evidenced the mycobacterial outer membrane (mycomembrane) by cryo-electron microscopy of vitrous section. He also developed a method to isolate it then deciphered its protein and lipid composition.
